The wolves approached a place in the Europa League with a 2-0 victory over Crystal Palace, while Sheffield United's European hopes were dashed by a 1-0 loss against Everton on Monday.
At the other end of the Premier League table, Brighton guaranteed their survival in the Premier League with a 0-0 draw against Newcastle. The wolves, now undefeated in three games, opened the scoring four minutes before the break in Molineux.
Andros Townsend lost possession and Joao Moutinho found Matt Doherty, who crossed for Portuguese midfielder Daniel Podence to head his first goal for the Lobos.
Jonny sentenced Palace to a seventh successive loss in the 68th minute, returning to shoot home from close range after Adama Traore and Raúl Jiménez opened Palace's defense.
Nuno Espirito Santo's team moved up to sixth place, one point above Tottenham, and will surely qualify for the Europa League if they win at Chelsea in their last game of the season on Sunday.
They would also qualify if Tottenham lose at the Palace in their final match.
"The idea for improvement and the construction process makes sense and today we achieved two more points than last season with one game to go," said Nuno.
"Sunday's game is very difficult, but let's prepare well and compete."
At Bramall Lane, Sheffield United received a second successive loss, ending their attempt to cap a magnificent first season in the top flight by winning a European spot.
Carlo Ancelotti's team won it in the 46th minute when Brazilian forward Richarlison was met by Gylfi Sigurdsson's free kick with a header for his 15th goal of the season.
Chris Wilder's eighth place is five points behind Wolves and four points behind Tottenham with one game left.
"I am disappointed by today's players. But what you are trying to do in one season is get your club through and we have done it in a ridiculous way," Wilder said.
"The top six should never have been in the equation from the moment we set foot in the division. There are a lot of clubs that would switch places with us."
The teams finishing fifth and sixth will surely make it to the Europa League next season.
If the FA Cup winners finish in the top six, seventh place would also qualify for the Europa League.
Arsenal, which faces Chelsea in third place in the FA Cup final, is currently in 10th place, meaning that Tottenham is in danger of a first season without competition in Europe from 2009-10.
